**Background**
Dipeptidyl peptidase IV (DPP-IV), also known as CD26, is a serine protease that plays a critical role in the metabolism of various bioactive peptides and hormones. One of its most significant physiological functions is the degradation of incretin hormones, such as glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP), which are essential for glucose homeostasis. Due to its pivotal role in regulating insulin secretion and blood glucose levels, DPP-IV has become a primary therapeutic target for the treatment of type 2 diabetes mellitus. Beyond metabolic regulation, DPP-IV is also involved in immune response and inflammatory processes. Consequently, developing precise tools to monitor and quantify the enzymatic activity of DPP-IV is essential for drug discovery and biochemical research. In this context, we will introduce a specific fluorescent substrate – Gly-Pro-AMC.
**Definition**
Gly-Pro-AMC hydrobromide is a fluorescent dye used as a specific substrate for the detection and measurement of Dipeptidyl peptidase IV (DPP-IV) activity. According to the Gly-Pro-AMC description, this compound serves as a sensitive probe that releases a fluorescent signal upon cleavage by the target enzyme.
**In Vitro Studies**
The Gly-Pro-AMC biological activity is characterized by its ability to be specifically hydrolyzed by DPP-IV, allowing for the quantification of enzyme kinetics. In vitro studies have demonstrated that Gly-Pro-AMC hydrobromide (50 μM; 20 min; 37 °C) can be effectively utilized as a specific fluorescent substrate for detecting DPP-IV activity. The detection of the released fluorophore is performed under specific fluorescence conditions, with an excitation wavelength of 350 nm and a fluorescence emission spectrum detection at 450 nm. This Gly-Pro-AMC protocol has been applied in various research settings, including the evaluation of DPP-IV inhibitory peptides derived from soybean, lupin, and Spanish dry-cured ham in human intestinal Caco-2 cells and ex vivo human serum. In conclusion, Gly-Pro-AMC is a highly specific fluorescent substrate for the detection of DPP-IV activity.
